WebOct 11, 2024 · We have reported that tumor necrosis factor (TNF)-related biomarkers, such as TNF receptors (TNFR1 and TNFR2) and progranulin (PGRN), are associated with the pathogenesis of obesity, diabetes, and CKD [ 14 – 16 ], and predict GFR loss and/or mortality in patients with diabetes and/or CKD [ 17 – 20 ]. TNF was thought to be produced primarily by macrophages, but it is produced also by a broad variety of cell types including lymphoid cells, mast cells, endothelial cells, cardiac myocytes, adipose tissue, fibroblasts, and neurons. Large amounts of TNF are released in response to lipopolysaccharide, other bacterial products, and interleukin-1 (IL-1). In the skin, mast cells appear to be the predominant source of pre-formed TNF, which can be released upon inflammatory sti…

WebFeb 1, 2024 · Sustained overexpression of tumor necrosis factor-α (TNF-α) appears to be one of several maladaptive mechanisms that contribute to the progression of heart failure. 3 TNF-α mRNA and protein are expressed in the failing human heart, but neither one is detectable in the healthy heart. 45678 TNF-α has pleiotropic effects that may accelerate … WebNov 6, 2024 · Circulating TNF was found to increase upon treatment, which may be explained by a prolonged TNF half-life, due to its tight binding to the antibody adalimumab, which has a very long half-life. As expected, TNF was biologically inactive, because the high amounts of TNF were essentially completely in complex with adalimumab.
